The specific steps are as follows:
1. Select 500g fresh carrots, wash them with clean water, drain and chop them, then dry them in an oven at 40 ℃ for about 2h, and further crush and screen the dried carrots. Note that the carrot must be thoroughly crushed.
2. Put the sample into a 500 ml round bottom flask, add 200 ml petroleum ether and mix well. Install the extraction reflux device as shown in Figure 6-7 in the textbook, extract for 30 minutes, and then filter the extract to remove solid substances.
3. Install distillation unit to concentrate the extracted samples.
4. Collect the sample in the receiver, observe the color and smell of the sample, and identify it by paper chromatography. Clean filter paper shall be selected during chromatography. In order to prevent pollution to the filter paper during operation, direct contact with the filter paper by hand shall be avoided as far as possible, and operation can be carried out with gloves. When sampling, it should be noted that the spot should not be too large (the diameter should be less than 0.5 cm). If it is dried with a hair dryer, the temperature should not be too high, otherwise the spot will turn yellow. Roll the filter paper with good samples into a cylindrical shape. When rolling the paper, pay attention to that both sides of the filter paper cannot contact each other, so as to avoid the accelerated movement of solvent along both sides of the filter paper due to capillary phenomenon, and the uneven front edge of solvent, which will affect the results.
